SIRPα （Signal regulatory protein α）

Signal regulatory protein α (SIRPα) is a regulatory glycosylated membrane protein from the SIRP family and is mainly expressed in bone marrow cells, stem cells, or nerve cells.

Signal regulatory protein α (SIRPα) is the most important member of the SIRP family. On the one hand, SIRPα can be activated by a variety of mitogens and phosphorylated, and binds to SHP-1 and SHP-2 through its ITIM domain to transmit inhibitory signals. On the other hand, SIRPα can also inhibit the activation of downstream pathways by interacting with CD47 ligands and transmit negative signals. SIRPα plays an important role in the regulation of innate immunity. In macrophages, LPS can induce down-regulation of SIRPα expression through transcriptional inhibition and protein degradation pathways. SIRPα inhibits the activation of MAPK, IKKs, NF-κB and IRF3 by blocking the effect of SHP-2. Reduce the release of inflammatory factors and IFNβ. Studies have shown that SIRPα in dendritic cells (DC) can also negatively regulate the functions of DC and T cells in both directions by binding to CD47 on the surface of T cells. On the one hand, it inhibits the expression of IL-12R by T cells and makes adult CD4+ and CD8 + T cells are less responsive to IL-12: on the other hand, they inhibit the phenotypic and functional maturation of immature DCs and inhibit their cytokine release.
